چکیده

An earlier report (1) dealing with the distribution of isotopic nitrogen in cells of Axotobacter vinelandii fixing molecular nitrogen provided evidence that ammonia might be the initial stable form of nitrogen in the fixation process. This conclusion was based on the observation that the N’s supplied this organism accumulated in the glutamic acid fraction, as had been already observed in higher plants (2) and animals (3) supplied ammonium labeled with the heavy nitrogen isotope. Comparable studies in which ammonium containing N15 was given to Azotobacter vinelandii are reported here.

Regulation of nitrogen fixation in Azotobacter vinelandii OP and in an apparently partially constitutive mutant.

Methylamine and 2-methylalanine appeared to act as co-repressors of nitrogenase in Azotobacter vinelandii OP. They inhibited the growth of this organism on molecular nitrogen but not on nitrate, ammonia, or Casamino Acids; they prevented the formation of nitrogenase by cells transferred from repression to induction conditions; and they did not inhibit the activity of nitrogenase in vitro. A mut...

Correlation of ultrastructure in Azotobacter vinelandii with nitrogen source for growth.

Azotobacter synthesizes an extensive internal membranous nework when grown with air (N(2)), i.e., under conditions when these bacteria fix nitrogen. Very slight quantities of internal membrane, concentrated mainly about the cell periphery, are formed when Azotobacter grows with fixed nitrogen, i.e., ammonia and amino acids. Compared to cells growing with ammonia, cells utilizing atmospheric nit...

Studies on Azotobacter Chroococcum Beij.

The study of the metabolism of Azotobacter has been generally approached from the standpoint of the r6le of this organism in the nitrogen cycle in Nature, but the fact that the ability to fix free nitrogen is regulated by the presence or absence of combined nitrogen in the medium has not been given serious attention although it is made plain in the works to be cited. Bejierink and Van Delden (1...

Molecular identification of Azotobacter isolated from the rhizosphere of Karunvariety of barley in desert and its comparison with standard Azotobacter andinvestigation of some of the vegetation indicators after inoculation

This study was conducted in acompletely randomized block design with three replications using three concentrations of 107, 105, and 103cfu/ml of Azotobacter isolated from the rhizosphere of Karun varieties of barley (Az. salentris) and standard Az.chrococom along with control (zero concentration of bacteria). The aim of this study was to study the reactions of barley plantin response to inocula...

The Utilization of Nitrogen in Hydroxylamine and Oximes by Azotobacter vinelandii.

Recent studies have provided several types of evidence supporting the view that ammonia is the key intermediate in nitrogen fixation by Azotobacter. An alternative hypothesis based on hydroxylamine depends almost entirely on the detection of traces of oximes in cultures fixing nitrogen. Although the two hypotheses are not mutually exclusive (Burris and Wilson, 1945), hydroxylamine remains an im...
